The Effect of Various Extraction Methods on Betanin Identification and Total Betalain Levels in Beets (Beta Vulgaris L.)

Beets (Beta vulgaris L.) are widely cultivated in various regions, including the Karo region, and are known for containing betalain, compounds that have pharmacological activity as antioxidants, hepatoprotectors, anti-inflammatory, and reduce oxidative stress. The betalain content in beets is a crucial factor in determining their potential health benefits. In this study, we aim to compare the content of betanin and total betalain obtained from three different extraction methods: maceration, reflux, and extraction with the help of microwave-assisted extraction. The method used in this study is an experimental study that involves the three extraction methods, accompanied by qualitative testing of betanin using thin layer chromatography and quantitative testing of betalain with ultraviolet-visible spectrophotometry.

Betalains are a group of water-soluble pigments that are responsible for the red, yellow, and orange colors of beets. They have been shown to have various pharmacological activities, including antioxidant, anti-inflammatory, and hepatoprotective effects. The betalain content in beets is influenced by various factors, including the extraction method used. In this study, we aim to investigate the effect of different extraction methods on the betanin content and total betalain levels in beets.

The study used three different extraction methods: maceration, reflux, and extraction with the help of microwave-assisted extraction. The beet tuber samples were obtained from the Karo region and were extracted using each of the three methods. The extracts were then subjected to qualitative testing using thin layer chromatography (TLC) and quantitative testing using ultraviolet-visible spectrophotometry (UV-Vis).

The results of the qualitative testing carried out by TLC show that all beet tuber extract samples are extracted by the method of maceration, reflux, and extraction with the help of microwaves containing betanin with an extract RF value of 0.487, which is close to the standard betanin value of 0.525. The total level of betalain in maceration extract, reflux, and extraction with the help of microwaves is 11.57 mg/g ± 0.28; 10.59 mg/g ± 0.14; and 11.50 mg/g ± 0.06, respectively. From this result, it can be concluded that the extraction method with the help of microwaves produces the highest amount of betalain.

The results of this study show that the extraction method has a significant impact on the total level of betalain in beets. The extraction method with the help of microwaves produces the highest amount of betalain, followed by maceration and reflux. This suggests that the selection of the right extraction method is crucial in obtaining optimal bioactive compounds from beets. The discovery of this study can be beneficial for the development of health products that utilize beet tubers as raw materials, especially in the food, pharmaceutical, and cosmetic industries.

In conclusion, this study confirms that the extraction method has a significant impact on the results obtained and can be a reference for further research in the exploration of bioactive compounds from other plants. The selection of the right extraction method is crucial in obtaining optimal bioactive compounds from beets. The discovery of this study can be beneficial for the development of health products that utilize beet tubers as raw materials.

The use of microwave-assisted extraction as the preferred method for extracting betalain from beets.
