Introduction

The genetic diversity of oil palm (Elaeis guineensis Jacq.) is a crucial aspect of its development, particularly in the face of environmental changes and disease attacks. The Moderate Ganoderma Moderate (MTG) variety of oil palm is one of the most widely cultivated species, and its genetic diversity is essential for the sustainability of the palm oil industry. In this study, we employed Random Amplified Polymorphic DNA (RAPD) markers to analyze the genetic diversity of individual MTG varieties.

Analysis of genetic diversity is a vital step in the development of superior plant varieties, especially for commodity plants such as oil palm. High genetic diversity not only helps in the restoration of plants from disease attacks but also increases plant resilience to environmental changes. In the case of oil palm, genetic diversity is very important to increase resistance to diseases such as ganoderma, which can result in significant losses in production. The genetic diversity of oil palm is influenced by various factors, including the selection of primary oligonucleotides, the number of DNA samples, and the software used for analysis.

Research Methodology

The RAPD method is a technique used to merge random segments from DNA, which allows researchers to evaluate the level of polymorphism and genetic relationships between individuals in the same species. The advantage of this method is that it is fast, simple, and does not require previous knowledge about the sequence of DNA from the species being studied. In this study, the exact selection of primary oligonucleotides is very influential on the results of the analysis obtained. The primary oligonucleotides used in this study were OPM-01, OPD-20, and SB-19.

Materials and Methods

This study was conducted at the Network Culture Laboratory of the Faculty of Agriculture, University of North Sumatra, from March 2017 to February 2018. A total of 27 DNA samples were used in this study, which were taken from stocks originating from PT Socfindo. The DNA samples were analyzed using the RAPD method, and the results were obtained using the Darwin 6.0.15 software.

Results

The results of the analysis showed that there were 16 identified DNA bands, with fragments varying between 191 bp to 1328 bp. The average percentage of polymorphism found is 94.3%, which shows that genetic diversity in this sample is quite high. Furthermore, to determine the genetic distance and form a dendogram, Darwin 6.0.15 software is used. The results of the analysis showed that the 27 samples were grouped in three clusters, with genetic distances ranging from 0.0 to 0.83, and total genetic diversity reached 51.73%.

Discussion

The results of this study indicate that the MTG variety of oil palm has a high enough genetic diversity. The formation of three clusters based on genetic similarities indicates the presence of genetic variations between the samples. Varied genetic distances from 0.0 to 0.83 indicate that although there are similarities between individuals, there are also significant differences that can be utilized in the breeding program. The total genetic diversity of 51.73% shows a great potential for the selection of superior individuals who can support the sustainability of oil palm production.

Conclusion

Through this study, it can be concluded that the moderate variety resistant to Ganoderma (MTG) from oil palm has a high enough genetic diversity. Data obtained from RAPD analysis can be used as a basis for the development of better varieties and more resistant to disease. Knowledge of genetic diversity is very important for future breeding strategies and can contribute to increasing productivity and sustainability of the palm oil industry.
