Introduction

Purple cabbage, a vegetable renowned for its vibrant color, is a rich source of essential nutrients, including water, protein, fat, carbohydrates, fiber, and minerals such as calcium, phosphorus, and iron. Additionally, it is rich in vitamins, sodium, potassium, cyanohydroxibutena, and anthocyanin, the compounds responsible for its distinctive purple hue. However, in the context of health, the safety of vegetables circulating in the community is often uncertain. Many of them are suspected of being contaminated with heavy metals, such as cadmium (CD) and lead (PB). Therefore, it is crucial to examine the heavy metal content in vegetables, including purple cabbage.

Background

Heavy metal contamination in vegetables is a significant concern, as it can pose a threat to human health. Cadmium and lead are two of the most common heavy metals found in vegetables, and their presence can lead to various health problems, including kidney damage, anemia, and even cancer. In this study, we aimed to identify the compounds contained in purple cabbage through chromatographic techniques and analyze the possibility of heavy metal contamination that can endanger health.

Methodology

The process of making the extract began with a purple cabbage simplicia maceration using 96% ethanol solvent for five days. The resulting maserat was then compacted using a rotary evaporator. The ethanol extract from purple cabbage (EKU) was then analyzed using paper chromatography (KKT) and thin layer chromatography (TLC) with a mobile phase variation. Additionally, the CD and Pb metal content was tested using the atomic absorption spectrophotometer (SSA).

Results

The results of the analysis using KKT showed that the most effective mobile phase was the combination of butanol, acetic acid, and water (BAA). In this test, four compounds were obtained with a rf value of 0.30; 0.39; 0.57; and 0.86 using NH reagent. Furthermore, using reagent fecl, two compounds with RF values of 0.18 and 0.48 were found, and with AlCl reagents that produced two compounds with RF values of 0.18 and 0.55.

The analysis using TLC showed that the combination of the n-hexane and ethilatate mobile phase with a ratio of 70:30 produced six compounds, of which four of them were terpenoid/steroid compounds that were purple with a value of RF 0.29; 0.69; 0.81; and 0.86. Furthermore, in the TLC experiment with the ratio of chloroform-menangol 90:10, seven saponin compounds were found. Of these seven compounds, four of them were blue with a RF value of 0.18; 0.27; 0.36; and 0.45, while three other compounds were violet with RF value of 0.75; 0.86; and 0.95.

Heavy Metal Content Analysis

From the results of the examination of heavy metal content, it was found that the cadmium content in the EKU was 0.0526675 ± 0.0082935 mg/kg, while the measured lead level was 0.1863148 ± 0.0386178 mg/kg. These results provide an important indication of the presence of heavy metals in purple cabbage, which can potentially endanger human health.

Conclusion

This study not only succeeded in identifying compounds in purple cabbage but also showed the need for tighter supervision of heavy metal contamination in vegetables. Given the high community interest in healthy vegetables, the results of this study function as a warning of the importance of food safety and potential health risks that may arise due to consumption of vegetables contaminated with heavy metals.
